Product Description

本制品是采用 SYBR Green I 嵌合荧光法进行 qPCR 的试剂盒，是一种 2X 预混液，反应液配制十分简单，仅需加入引物、模板及 RNase free water 即可进行 qPCR 反应，使用方便。本制品是在 SYBR Green Premix Pro Taq HS qPCR Kit II（Code. AG11702）产品基础上预混了高浓度 ROX 的 2X Premix 试剂，无需额外添加 ROX，方便使用（反应中 ROX的终浓度为 0.4 μM）。
本制品中 SYBR Green I 浓度、PCR 反应体系都进行了优化，采用了反应性能优越的 Pro Taq HS 体系，其混合了能在常温下抑制 Taq 酶活性的单克隆抗体，从而可以有效抑制非特异性扩增，提高 PCR 扩增效率，同时能够在宽广的定量区域内获得良好的工作曲线，对靶基因进行准确定量检测。

 

Product Advantages

② The product is optimized for SYBR Green I concentration and PCR reaction system, which is characterized by high amplification efficiency and strong amplification specificity.
③ This product is premixed with ROX Reference Dye, which is convenient to use and effectively reduces the experimental error caused by single addition of ROX. It is suitable for high concentration ROX calibrated quantitative PCR instruments or quantitative PCR instruments that do not need to be calibrated.

Example 1

采用本试剂盒进行荧光定量检测 Mouse GAPDH Gene（GC 含量 56%），cDNA 模板添加量（相当于 Total RNA 量）为 10 ng ~ 100 fg。cDNA 的合成使用本公司的 Evo M-MLV 反转录预混型试剂盒 (含去除 gDNA 试剂，用于qPCR) (Code. AG11728)。
Quantification instrument used: Bio-Rad CFX96TM Real-Time PCR Detection Systems（无需 ROX 校准）。
结果如下：

The results are shown above:
① Working curve R2=0.9992，扩增效率93.5%。
② 可以在宽广的模板范围内进行准确的定量，10 ng ~ 100 fg cDNA浓度（相当于 Total RNA 量）范围内扩增曲线呈现良好的线性关系。
③ The melting curve has a single peak type, no spurious peaks, and strong amplification specificity.

Example 2

采用本试剂盒进行荧光定量检测 Mouse GAPDH Gene (GC 含量 56%)，cDNA 模板添加量（相当于 Total RNA 量）为 10 ng ~ 100 fg。cDNA 的合成使用本公司的 Evo M-MLV 反转录预混型试剂盒(含去除 gDNA 试剂，用于 qPCR) (Code. AG11728)。
所用定量仪器：ABI StepOne Real-Time PCR System（需 ROX 校准）。结果如下：

The results are shown above:
①Working curve R2=0.9995，扩增效率 95.7%。
②Accurate quantification can be performed over a wide template range, with amplification curves showing good linearity over the range of 10 ng ~ 100 fg cDNA concentration (equivalent to Total RNA amount).
③ The melting curve has a single peak type, no spurious peaks, and strong amplification specificity.
