DNA Ligation Kit Ⅲ Ver.2 is a simple, fast and efficient targeted seamless cloning kit, which can quickly and efficiently clone one or more DNA fragments into any position of any vector in a targeted and seamless manner without being restricted by the enzyme cut site. As a new generation of seamless cloning kit, this product has a wide range of applications and can efficiently connect different sizes of vectors and insert fragments, especially suitable for multi-fragment seamless cloning.
The DNA ligation reaction is based on OK, Clon. Enzyme II catalyzes the accurate and efficient insertion of fragments containing sequences (15 ~ 20 bp) that are consistent with both ends of the vector into the linearized vector. When designing the PCR primer for the target fragment, it is only necessary to add the sequence complementary to the 5’ end of the vector to the 5’ end of the primer, and then amplify the target fragment through the primer, and then insert the target fragment into the vector without the need for restriction enzyme digestion or phosphorylation of the PCR fragment.
