This product is a rapid and highly sensitive test kit based on XTT [2,3-di-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazole-5-carboxanilide], which is widely used in cell proliferation and cytotoxicity. The detection principle is that XTT can be reduced to water-soluble orange-yellow dirty product by reductive dehydrogenase generated in living cells under the action of electron coupling agent 1-methoxy PMS, and the absorbance can be detected at 450 nm using an enzyme marker, and the stronger the cell viability, the more dirty is generated and the darker the color is, and the absorbance is directly proportional to the cell viability within a certain range, so cell proliferation and toxicity analysis can be carried out by XTT. Therefore, XTT can be used to analyze cell proliferation and toxicity. This product is easy to operate, only need to add the working solution prepared by XTT detection solution and 1-methoxy PMS directly into the cells, incubated at 37℃ for a certain period of time; there is no need to collect the cells, no need to wash the cells, and the absorbance can be measured at 450 nm using an enzyme marker directly.
This product can be widely used in cell proliferation assays (e.g., cell proliferation induced by growth factors, cytokines, etc.); cytotoxicity assays (e.g., cytotoxicity of anticancer drugs); and so on.
1. This product is easy to operate, first mix XTT Reagent and Electron Coupling Reagent in the ratio of 100:1 to form a working solution (ready to use), mix well and can be used; the working solution can be added directly to the cell culture medium, without the need to collect cells or wash cells.
2. This product is non-toxic to cells, cells treated with XTT working solution can be added to cell culture medium after discarding the supernatant for further cultivation. At the same time, after adding XTT working solution, it can be continuously detected by enzyme marker at different time points and analyzed at multiple time points, which also makes the detection time more flexible.
3. High detection sensitivity and wide linear range, e.g., 0~1 X 10 using 96-well plate.5 Cell range shows good linearity.
4. Fast detection speed, short reaction time, the test results can be obtained within 1~4 h.
5. Phenol red and serum have no effect on the XTT test.
