Product Description

This kit is a combination of ARMS-PCR and SYBR Green I chimeric fluorescence method for SNP genotyping. It is a 2X pre-mixed reagent that requires only primers, template, and RNase free water, making the preparation of the reaction solution very simple. The product is optimized for SYBR Green I concentration and PCR reaction system, which can amplify templates of different complexity; the product uses modified Taq DNA Polymerase, which has high amplification efficiency and sensitivity, and monoclonal antibody that can inhibit the activity of DNA Polymerase at room temperature has been added. At the same time, a monoclonal antibody that can inhibit the activity of DNA Polymerase at room temperature is also added, which can effectively inhibit the formation of primer dimer and non-specific amplification, and it can carry out highly sensitive Real Time PCR reaction, which is very suitable for the detection of SNP genotyping.

Example 1

Use this product for fluorescence quantitative PCR in mice. GAPDH genes, the template was added at 100 ng - 1 pg, and cDNA was synthesized using the company's Evo M-MLV Reverse Transcription Premix Kit (with gDNA removal reagent for qPCR) Ver.2 (Code No. AG11728). The quantification instruments used were ABI QuantStudioTM 5 Real-Time PCR Systems. the results are shown below:

1. Working curve R2= 0.999, amplification efficiency 100%;
2. Accurate quantification is possible over a wide template range, with good linearity in the amplification curve over the 100 ng - 1 pg concentration range;
3. The melting curve has a single peak type, no spurious peaks, and strong amplification specificity.

Example 2

ABI QuantStudio is used for the quantification of different genotypes (wild type, mutant) of different samples.TM 5 Real-Time PCR Systems, different types of templates can be accurately differentiated by comparing the differences in Ct values, and the results are as follows:

The results are shown above:
1. In the typing result 1, according to the amplification curve, it was found that the wild-type template amplified normally, and the mutant template did not have peaks, which could distinguish the mutant type from the wild-type significantly;
2. In typing result 2, the difference between mutant and wild-type Ct was found to be ≥ 14 according to the amplification curve, which can significantly distinguish the mutant from the wild-type.
