Product Description

This kit is based on the JC-10 fluorescent probe and can rapidly and sensitively detect changes in cellular mitochondrial membrane potential, which can be used for the detection of early apoptosis.
JC-10 is a water-soluble fluorescent probe widely used to detect the mitochondrial membrane potential ΔΨm, which accumulates in mitochondria in a potential-dependent manner. In normal mitochondria, where the mitochondrial membrane potential is high, JC-10 aggregates in the mitochondrial matrix to form a polymer, emitting intense red fluorescence; in unhealthy mitochondria, due to the decrease or loss of mitochondrial membrane potential, JC-10 exists in the cytosol as a monomer, producing green fluorescence. Therefore, changes in the emission wavelength of JC-10 fluorescence can be detected by fluorescence microscopy or flow cytometry, reflecting changes in mitochondrial membrane potential.
Loss of mitochondrial membrane potential is a hallmark event in the early onset of apoptosis. When cells are stimulated by apoptotic signals, the intracellular mitochondrial membrane potential is altered and membrane permeability is increased, resulting in the release of mitochondrial proteins, including cytochrome C, from the mitochondrial matrix into the cytosol. The release of cytochrome C is accompanied by a complete loss of membrane potential, which triggers a cascade of apoptotic responses.
CCCP (Carbonyl cyanide 3-chlorophenylhydrazone) is a potent mitochondrial oxidative phosphorylation uncoupler that induces the mitochondrial inner membrane to H+ Produces permeability, leading to a loss of membrane potential in mitochondria. This product provides CCCP as a positive control for inducing a decrease in mitochondrial membrane potential.

test case

Jurkat cells treated with CCCP for 15 min and untreated cells were stained with JC-10 (final concentration 2 μM) and observed using flow cytometry and fluorescence microscope, the results are shown below, the CCCP treated cells appeared in green color and the normal cells appeared in red color.
