Product Description
This product is designed to be used in RNA libraries containing UMI (Unique Molecular Identifier) sequences with 5 ’ TSO-joined primers, specifically designed to be used with the AccuNext Strand-specific RNA Library Preparation Kit (Illumina) (Code No. AG12503 / AG12504) and AccuNext The 5’-Adapter Primer Mix can be completely replaced with this product without any change in usage for use with the Chain-Specific Single Cell/Micro RNA Construction Kit (Illumina) (Code No. AG12526/AG12527).
The UMI sequence of this product is a short random sequence of 8 nt, which can be used to add a unique molecular barcode to each cDNA during reverse transcription, and specifically label each cDNA; after constructing the library by PCR amplification and sequencing, the reads obtained from sequencing can be filtered out the duplicated reads and the sequences with errors in PCR amplification with high accuracy by the bioinformatics software. Therefore, the use of UMI-conjugated primers for library construction and sequencing can reduce PCR bias, exclude false positives, improve sequencing accuracy, and provide more accurate information for sequencing analysis.
