Product Description

This is a colorimetric protein quantification kit based on the BCA method (Bicinchoninic Acid), which is based on the principle that in an alkaline environment, the peptide bonds in proteins can reduce Cu²⁺ to Cu⁺, and the resulting Cu⁺ reacts with the BCA reagent to form a stable blue-purple The resulting Cu⁺ reacts with the BCA reagent to form a stable blue-violet complex with strong absorption at 562 nm, and its absorbance value shows a good linear relationship with the protein concentration.
There are two modes of measurement available: standard concentration protein measurement (standard mode) (50 ~ 2000 μg/ml) and micro concentration protein measurement (micro mode) (<50 μg/ml), so please select the appropriate mode according to your experimental needs.
This product can be used in microplate and cuvette method for detection, microplate method is easy and fast to operate, only a small amount of protein samples; cuvette method requires a relatively large number of protein samples, cuvette method is longer than the optical diameter of the microplate, so the sensitivity will be better than the microplate method.
This product is highly tolerant to interfering substances such as surfactant SDS, Triton X-100, Tween-20, etc. in protein samples, but it is not suitable for quantification of protein samples containing reducing agents or chelating agents. The maximum tolerated concentrations of different interfering substances in protein samples are shown in Appendix 1.

*1: BCA Reagent A and BCA Reagent B are packaged in Package 2-1 and stored at room temperature (15~30℃), and BSA Standard Solution (2 mg/ml) is packaged in Package 2-2 and stored at 4℃.
*2: Using the microplate method (200 μl reaction system), this product can perform 500 reactions.

Preservation and transportation

Preservation Temperature:

Example 1

Different BSA concentrations were determined using the standard assay mode of the microtiter plate method (assay range: 250 ~ 2000 μg/ml) (reaction conditions: 37°C, 30 min) and plotted in a standardized curve, which is shown below:

The results are shown in the figure above: the BSA standard assay mode was used for the BSA standard curve assay, and BSA showed a good linear relationship in the concentration range of 250 ~ 2000 μg/ml.

 

Example 2

Different BSA concentrations were determined using the standard assay mode of the cuvette method (assay range: 250 ~ 2000 μg/ml) (reaction conditions: 37°C, 30 min) and plotted in a standardized curve, which is shown below:

The results are shown in the figure above: the BSA standard curve was determined using the standard assay mode of the cuvette method, and BSA showed a good linear relationship in the concentration range of 250 ~ 2000 μg/ml.

 

Example 3

Different BSA concentrations were determined using the standard assay mode of the microtiter plate method (assay range: 50 ~ 250 μg/ml) (reaction conditions: 60°C, 60 min) and plotted in a standardized curve, which is shown below:

Using the standard assay mode of the microtiter plate method for BSA calibration curve assay, BSA showed a good linear relationship in the concentration range of 50 ~ 250 μg/ml.

 

Example 4

Different BSA concentrations were determined using microplate assay in micro assay mode (assay range: 0 ~ 50 μg/ml) (reaction conditions: 60°C, 60 min) and plotted in a calibration curve, which is shown below:

The BSA calibration curve assay was performed using the microtiter plate method in micro assay mode, and BSA showed a good linear relationship over the concentration range of 0 ~ 50 μg/ml.
