Product Description

This product is ready-to-use Pro Taq HS enzyme PCR reaction 2x premix. For the PCR reaction, only the template and primers are added to the premix for amplification. This premix is easy to use, minimizes human error, and provides results in a short period of time. The Pro Taq The enzyme was developed by utilizing the principle of LA PCR in the company's high-performing Accurate Taq The enzyme has a high fidelity enzyme with 3′ → 5′ Exonuclease activity (Proof reading activity), which is ideal for amplification of DNA fragments over 10 kb and has good fidelity. The Taq monoclonal antibody is also included in the system for Hot Start PCR, where the antibody binds to the Taq enzyme before the reaction starts and inhibits its activity, thus suppressing non-specific amplification caused by primer annealing or primer dimerization at low temperatures. The antibody is inactivated during the initial DNA denaturation step when the PCR reaction is initiated, so it can be used under routine PCR reaction conditions. Most PCR products obtained with this product have an A base at the 3’ end and can be cloned directly into T vectors.

② 2X Pro Taq HS PCR Master Mix Ver.2 is thermally stable and can amplify ~40 kb of DNA fragments using λDNA as template and ~17.5 kb of DNA fragments using Human Genomic DNA as template.
③ A monoclonal antibody capable of inhibiting DNA polymerase activity has been added to this product, which can be used for hot start reaction. Before the PCR reaction, the antibody binds to the enzyme and inhibits the activity of DNA polymerase, effectively inhibiting the formation of primer dimers and non-specific amplification.

Example 1

Using λDNA as a template, a 40 kb DNA fragment can be amplified very well.

Human gDNA was used as a template, and a 17.5 kb DNA fragment was well amplified.

The gDNA of pig blood was used as a template to amplify 10 kb and 15 kb DNA fragments, which were able to amplify the target fragments.

The 10 kb and 15 kb DNA fragments were amplified using rice gDNA as a template, and the target fragments were well amplified.

Human gDNA was used as a template and different template amounts (500 ng, 100 ng, 10 ng, 1 ng) were added to amplify 10 kb DNA fragments, and the target fragments could be amplified with template amounts as low as 1 ng.
