AccuLyo Taq HS Premixed Lyophilizable Probe-based qPCR Kit (for ASFV, with UNG, without primer probe)

This product is a 2X Premix with a lyophilization protectant for the detection of African Swine Fever Virus (ASFV) by the probe method. It can be used directly for lyophilization, or can be lyophilized after addition of primers and probes. The lyophilized product is easy to store and transport at room temperature.
This product contains the superior reaction performance of Accurate Taq HS DNA polymerase, together with carefully optimized reaction buffer, improves PCR amplification performance and sensitivity of low concentration template detection, enabling highly sensitive Real Time qPCR reactions for accurate quantitative detection of target genes.
The dUTP/UNG anti-contamination system is introduced in this product. dUTP replaces dTTP in the PCR reaction, and the UNG enzyme selectively hydrolyzes dU-containing DNA strands without affecting non-dU-containing DNA strands, removing dU-containing contaminating templates introduced during the preparation of the PCR reaction system, thus effectively preventing the generation of false-positive results in PCR; At the same time, monoclonal antibody that can inhibit the activity of DNA Polymerase at room temperature is added to this product, which can effectively inhibit non-specific amplification, improve the sensitivity of the reaction and enhance the accuracy of the results.

Using African swine fever B646L standard material [ GBW ( E ) 091034 ] as template, the gene was detected by qPCR with fluorescent probe on both lyophilized and untreated lyophilized products placed at 37 ℃ for 30 days, and the amount of template added to the 20 μl reaction system was 30 copies and 0 copies (negative control). The performance of the lyophilized product placed at 37℃ for 30 days (green curve) was comparable to that of the untreated lyophilized product (red curve).
The quantitative instruments used for the assay were ABI QuantStudio^TM 5 Real-Time PCR Systems. the results of the assay are shown below:

The results are shown above:
1. For low-copy viral templates, the detection rate of lyophilized product placed at 37℃ for 30 days was 100%, which is comparable to that of untreated product.
2. Negative controls were not detected within 45 cycles.
