CytoDet Cell Viability Assay Kit (CCK-8)

This product is a rapid and highly sensitive test kit for cell proliferation and cytotoxicity based on WST-8 [2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl )-2H-tetrazole monosodium salt]. The detection principle is that WST-8 can be reduced to water-soluble orange-yellow dirty product by reductive dehydrogenase generated in living cells under the action of electron coupling agent 1-methoxy PMS, and the absorbance can be detected at 450 nm wavelength using an enzyme marker, and the stronger the cell viability the more dirty product is generated, and the darker the color is, within a certain range, the absorbance is directly proportional to the cell viability, so it can be analyzed for cell proliferation and cytotoxicity by WST-8. Therefore, WST-8 can be used to analyze cell proliferation and toxicity. This product is simple to operate, just add CCK-8 solution directly into the cells, incubate at 37 ℃ for a certain time, without collecting cells, without washing the cells, directly using the enzyme marker at 450 nm wavelength absorbance can be measured. The product is non-toxic to the cells, so the supernatant can be discarded and added to the cell culture medium for further cultivation after the experiment.
This product can be widely used in cell proliferation assays (e.g., cell proliferation induced by growth factors, cytokines, etc.); cytotoxicity assays (e.g., cytotoxicity of anticancer drugs); and so on.

1. It is a ready-to-use CCK-8 reagent, no need to prepare other solutions, can be used after mixing; the solution can be added directly to the cell culture medium, without the need to collect the cells and wash the cells.
2. This product has no obvious toxicity to cells, and the cells treated with CCK-8 Solution can be further cultured by adding it to the cell culture medium after discarding the supernatant. At the same time, after adding CCK-8 Solution, it can be continuously detected by enzyme marker at different time points and analyzed at multiple time points, which also makes the detection time more flexible.

4. Fast detection, short reaction time, within 1~4 h can be tested to obtain results.
5. Phenol red and serum have no effect on the CCK-8 Solution assay.

HL60 cells and 293T cells were inoculated into 96-well cell culture plates at different numbers (0, 2000, 4000, 8000, 16000, 32000, 64000, 100000 Cells) according to 100 μl of culture medium per well, and 20 μl of CCK-8 Solution was added to each well, and the absorbance was measured at 450 nm after incubation at 37℃ for 3 h. The results are shown below. The absorbance was measured at 450 nm, and the results, such as the figure below, showed a good linear relationship in the range of 0-100000 cells.
