This product is a high sensitivity test for cell proliferation and cytotoxicity based on MTT [3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyltetrazolium bromide]. The detection principle is that MTT can be reduced to blue-violet insoluble dirty products by the reductive dehydrogenase generated in living cells, and the absorbance can be detected at 570 nm using an enzyme marker, the stronger the cell viability, the more dirty products are generated, and the darker the color is, within a certain range, the absorbance is directly proportional to the cell viability, so it can be used to conduct the analysis of cell proliferation and toxicity by MTT. The operation of this product is simple, only need to add the MTT reagent directly into the cells, incubate at 37 ℃ for 3 h, then add the solubilization reagent, incubate at 37 ℃ for a certain period of time, no need to collect the cells, no need to wash the cells, and then directly use the enzyme marker to measure the absorbance at 570 nm wavelength.
This product can be widely used in cell proliferation assays (e.g., cell proliferation induced by growth factors, cytokines, etc.); cytotoxicity assays (e.g., cytotoxicity of anticancer drugs); and so on.
1. This product is easy to operate, only need to add MTT Reagent to the cell culture medium and incubate for a certain time, then add MTT Solvent Solution and incubate; the reaction solution can be directly added to the cell culture medium, without the need to collect cells and wash cells.
2. High sensitivity and wide linear range, e.g., 0~1X10 using a 96-well plate.5 Cell range shows good linearity.
3. This product has a wide range of applications and is suitable for viability testing of a wide range of adherent cells as well as suspension cells.
4. Phenol red and serum had no effect on the MTT assay.
