Product Description

DAPI is a fluorescent dye based on DAPI (4′, 6-diamidino-2-phenylindole), which is widely used for DNA and nucleus staining in cellular imaging techniques. The detection principle is that DAPI binds to the minor groove in the DNA double strand and emits an intense blue fluorescence (maximum excitation and emission of Ex = 364 nm and Em = 454 nm, respectively); DAPI itself has a low fluorescence background, but binding to dsDNA enhances the fluorescence intensity by about 20 times. The product is easy to operate, only need to dilute DAPI Solution to the appropriate concentration, add it to the sample, incubate for a certain period of time, then bright blue fluorescence can be observed, and can be observed by fluorescence microscope or flow cytometry.
DAPI is widely used for nuclear staining, apoptosis, and cell cycle analysis of living cells, fixed cells or tissues, and paraffin sections. The concentration of this product is 1 mg/ml and can be diluted to the desired working concentration, generally recommended working concentration of DAPI is 0.5~10 μg/ml. In addition, we also provide ready-to-use products CytoDet DAPI Staining Solution (Ready-to-Use) (Code No. AG51014 / AG51016), which is optimized to be used directly and does not need to be diluted. The concentration is optimized for direct use without the need for dilution.

Product Advantages

The working concentration of DAPI is recommended to be 0.5~10 μg/ml when used for staining of fixed cell nuclei.

Experiment example 1

The products were fixed with appropriate amount of 4% paraformaldehyde fixative at room temperature for 1×106 The cell nuclei showed blue fluorescence as shown in the figure below. 3T3-L1 cells in 12-well plates were washed twice with appropriate amount of PBS after removing the fixative, and then stained with 5 μg / ml DAPI staining working solution.

Example 2

Paraffin sections of mouse intestine were stained using 5 μg / ml of DAPI staining working solution, and the results were as shown below, with nuclei showing blue fluorescence.
