Product Description

Hoechst 33342 is a fluorescent dye based on Hoechst 33342 [2′-(4-ethoxyphenyl)-5-(4-methyl-1-piperazinyl)-2,5′-bis-1H-benzimidazole trihydrochloride], which is widely used in cellular imaging for the staining of DNA and the nucleus of cells. The detection principle is that Hoechst 33342 penetrates the cell membrane and binds to the adenine-thymine (A-T) groove of dsDNA to stain the nucleus. Hoechst 33342 has a low fluorescence background and emits an intense blue fluorescence when bound to dsDNA (maximum excitation and emission of Ex = 350 nm and Em = 461 nm, respectively). Hoechst 33342 staining is commonly used for apoptosis detection, which can be visualized by fluorescence microscopy or flow cytometry.Hoechst 33342 is also commonly used for general nuclear staining, or routine DNA staining. Hoechst 33342 is easy to use, simply dilute Hoechst 33342 solution to the appropriate concentration, cover the sample and incubate for a certain period of time to observe the bright blue fluorescence.
This product can be widely used for nuclear staining of living cells, fixed cells or tissues, paraffin sections as well as apoptosis and cell cycle analysis.

Product Advantages

1. This product is a ready-to-use dye, easy to operate, when staining fixed cells or paraffin section samples, it can be used to dilute it to the working concentration (0.5~10 μg / ml) with PBS or cell culture medium, add it to the cells, incubate at room temperature for a certain period of time for observation; when staining live cells, the solution can be added directly to the cell culture medium according to a certain ratio, without the need to collect the cells or wash the cells, and incubate for a certain period of time at 37℃ for observation. Experiment example 1

Hela cells were inoculated in 12-well cell culture plates one day in advance and stained with 10 μg/ml of Hoechst 33342 staining workup 24 h later, and the results are shown below, with nuclei showing blue fluorescence.

Example 2

1 ml of Hoechst 33342 staining work solution (10 μg / ml) was used to stain 1 × 106 Jurkat live cells were incubated at 37°C for 10 min, and the results were as shown in the figure below, the live cells showed light blue fluorescence, and some of the early apoptotic cells contained therein showed bright blue color (red arrows point to normal live cells, white arrows point to apoptotic cells).
