Product Description

After drinking alcohol, ethanol is oxidized to acetaldehyde in the liver by the enzyme ethanol dehydrogenase (ADH), and acetaldehyde is further converted to harmless acetic acid as a substrate catalyzed by the enzyme acetaldehyde dehydrogenase 2 (ALDH2); ALDH2 is a key enzyme for alcohol metabolism in the human body, and a mutation in the ALDH2 gene has a common single-nucleotide polymorphism (SNP) site (rs671), which reduces the body's ability to detoxify acetaldehyde. Mutations in the ALDH2 gene reduce the body's ability to detoxify acetaldehyde and may increase the risk of diseases such as cancer. Therefore, SNP testing of the ALDH2 gene can be used to analyze the ability to metabolize alcohol by examining the type of ALDH2 mutation (wild-type, heterozygous, or mutant), and to guide individualized alcohol consumption to prevent disease.
This product is based on TaqMan MGB method and specially developed for ALDH2 gene, the 12.5X GenDiff SNP Primer & Probe Mix for ALDH2 Genotyping in the kit contains two different fluorescent probes, FAM and VIC, FAM fluorescent channel is used for detecting wild type (G type), and VIC fluorescent channel is used for detecting mutant (A type), which can be used with 2X GenDiff SNP qPCR Mix for ALDH2 Genotyping and Lysis Buffer in the kit. The FAM fluorescence channel is used for detecting wild type (G type) and the VIC fluorescence channel is used for detecting mutant type (A type), and it can be used with the 2X GenDiff SNP qPCR Mix for ALDH2 Genotyping and the Lysis Buffer in the kit; this product is easy to operate, and you can observe the presence or absence of the two fluorescence probes after you have scraped the oral epithelium and lysed them into Lysis Buffer for quantitative amplification. The genotyping results can be obtained by observing the presence or absence of two types of fluorescent probes. The product is characterized by easy operation, high sensitivity, high reproducibility and strong anti-interference.

Product Advantages

① The product is simple to operate, oral epithelial cells can be sampled directly after quantitative amplification, by observing the presence or absence of the two fluorescent signals can be determined by the results, with the advantages of simple operation, simple analysis and reliable results.
② This product is characterized by high sensitivity, high differentiation, high repeatability and strong anti-interference.

Example 1

Oral epithelial cells were sampled from different populations, and different genotypes were detected using this product. The quantitative instrument used: Bio-Rad CFX96, and the judgment criteria and example charts are as follows:

1) Criteria for determination*1:

2) Example genotyping chart:

The results are shown above:

1) Only FAM fluorescence is detected as wild-type with high alcohol tolerance;
2) Those detected by both FAM and VIC fluorescence are heterozygous and have average alcohol tolerance;
3) Only VIC fluorescence was detected as mutant with weak alcohol tolerance.
