Product Description
This product is a transposase DNA library kit developed for illumina's high-throughput sequencing platform, which can prepare a sequencing library from a purified 1 ng DNA sample. Using a novel transposase and optimized buffer system, the product can complete DNA fragmentation and junction ligation in only 10 min, which is much simpler and more convenient than the traditional library preparation method, eliminating the need for cumbersome DNA fragmentation, end repair and junction ligation reactions, and significantly shortening the time for library construction. Since Transposome Mix V1 is very sensitive to the amount of DNA added, the exact amount of DNA added is critical to the success of the experiment, so it is recommended to add 1 ng of DNA template without adjustment. For other template incorporation amounts, 2 other kits with different template starting amounts are available, which are suitable for DNA template incorporation of 50~100 ng (CodeNo. AG12518/AG12519) and 5 ng (CodeNo. AG12522/AG12523), respectively.
The reaction system of this product has been carefully optimized, all reagents in the experiment should be used as provided in this product, it is not recommended to change the dosage and concentration of any reaction components or to replace the components in this product with other equivalent products in order to avoid obtaining bad results. If replacement is needed, please verify it first.

50 ng of HL60 Cell gDNA was used as a template, with the AccuNext Fast Tagment DNA Library Prep Kit for Illumina Ver.2 (50~100 ng Input DNA) (Code No. AG12518, 8 cycles of PCR amplification) was used to prepare the DNA library. DNA libraries were prepared using the MagSpherix DNA Beads (Code No. AG12547), were purified using 1.0X magnetic beads, length-sorted using 0.9X / 0.2X, 0.8X / 0.2X, 0.7X / 0.2X bead ratios, and then assayed using an Agilent 2100 Bioanalyzer. The results are shown in Figure 4 below.
